基本信息

BioNMRLab隸屬於中國科學技術大學生命科學學院和合肥微尺度物質科學國家實驗室,地址位於安徽省合肥市蜀山區黃山路445號,中國科學技術大學生命科學院。實驗室現有(截至2019年9月4日)中國科學院院士1名,正教授(不含院士)3名,副教授2名,講師、管理員、工程師、實驗員等6名,博士後3名,博士、碩士研究生30餘名。實驗室成立近30年間,培養了十餘名教授、二十餘名副教授、上百名碩士博士生等科研界人才,以及眾多行政領域、商業領域人才。

施蘊渝

施蘊渝,實驗室科研領軍人。分子生物物理學家,中國科學院院士第三世界科學院院士中國科學技術大學教授、博士生導師。
研究方向:蛋白質分子設計及藥物設計有關的基礎理論和方法學的研究,包括底物藥物與靶標分子結合自由能的計算,蛋白質穩定性的計算機模擬,蛋白質分子的隨機動力學模擬,蛋白質靜電相互作用研究,以及酶作用機理的計算機模擬。生物大分子蛋白質及核酸晶體學核磁共振波譜學結構與功能機理研究。

研究方向

實驗室主要套用核磁共振波譜學(NMR)綜合X-射線晶體學衍射,冷凍電鏡,生物分子生物學細胞生物學等方法來研究生物大分子結構動力學,蛋白質-蛋白質相互作用,蛋白質-DNA/RNA相互作用蛋白質-配體相互作用, 來解析生命過程中重要的生物大分子及其複合物的功能。

表觀遺傳結構生物學研究

染色質的結構是高度動態可塑的。在真核生物基因轉錄DNA複製DNA損傷修復過程中,染色質的高級結構要有序地受到外部因子如染色質重塑複合物,DNA及組蛋白修飾酶,組蛋白分子伴侶等外部因子的調控。同時核小體也會組裝與去組裝,並伴隨不同組蛋白變體的交換。這個新興領域目前仍有大量的科學問題懸而未決。我們因此綜合運用核磁共振和X-射線晶體衍射技術,在結構的基礎上闡明核小體翻譯後修飾及組裝的分子機制。<a 收縮。

非編碼RNA的結構生物學研究

轉運RNAtRNA)和核糖體RNArRNA)到snoRNA、microRNA、siRNAsnRNA、exRNA、piRNA、和lncRNA等,非編碼RNA是一類不會翻譯成蛋白質但卻具有重要生物學功能的RNA分子,在體內廣泛存在。最新的測序研究證實,人類基因組中約有98%的部分是不編碼蛋白的基因,而只一點與其他物種相比時有著鮮明的區別,一定程度上解釋了為什麼哺乳動物遠比其他物種複雜。
然而儘管非編碼RNA已被發現在包括基因表達調控剪下等細胞內過程中,以及癌症疾病的發生髮展中扮演重要角色,大量非編碼RNA的精確的功能意義以及它們的結構功能關係仍然很大程度上未知。目前的研究發現,RNA分子在結構和功能上存在很大的靈活性。另外,由於RNA分子帶有強負電,不易於結晶,使得使用X-射線晶體衍射方法對其進行結構研究變得困難。因此使用包括溶液核磁共振、小角散射、和冷凍電鏡等在內的其他結構生物學手段研究RNA結構變得極為重要。其中核磁共振在研究非編碼RNA這類結構上存在高度靈活性的生物分子時具有一定的優勢。
實驗室目前主要使用核磁共振和X-射線晶體衍射方法研究大腸桿菌里的非編碼RNA以及它們與RNA分子伴侶蛋白Hfq複合物的結構。我們希望通過這些研究能進一步揭示非編碼RNA在應激條件下調控下游mRNA翻譯表達的分子機制。

生物大分子核磁共振方法研究

X-射線晶體學核磁共振波譜學冷凍電鏡是目前解析高分辨生物大分子三維結構的主要方法。其它方法,包括X-射線小角散射,電子順磁共振(EPR),PCS和順磁弛豫增強(PRE)等方法提供了重要的補充信息.我們套用和發展上述多種結構解析方法,使用多種解析度下的實驗室數據和分子動力學建模來研究生物大分子在多種時間尺度下的結構和動力學性質。

基於片段的小分子化合物篩選

靶標蛋白-蛋白相互作用體系被譽為藥物發現的明珠。選擇性的阻滯特定蛋白-蛋白相互作用極大地擴充了現有的藥物靶點。蛋白-蛋白相互作用的熱點區域相比於經典的激酶口袋,更為狹窄舒緩,因此發現高親和力、高選擇性的小分子抑制劑面臨著巨大的挑戰。2000年以來興起的基於片段的先導化合物發現在新型的靶點研究上成果豐碩,目前已有一個藥物上市,數十種藥物進入臨床。我們建立了國內首批高度自動化的核磁片段篩選平台,針對和腫瘤增殖和遷移密切關聯的表觀遺傳靶點和小GTP酶體系,發現了這類靶點的弱結合的苗頭化合物,並在結構指導下拼裝為類似先導化合物的化學探針。探索這類探針在細胞內阻滯相應的蛋白-蛋白互作,從而抑制腫瘤增殖和遷移的分子機制。
