項目摘要
肌萎縮側索硬化症（ALS）是一種累及脊髓前角細胞、腦幹運動神經核及錐體束，具有上、下運動神經元損害並存的致死性神經變性病，確切發病機制不清，能量代謝異常是其發生與發展的重要病理生理機制。本課題前期實驗發現AMPK/SIRT1能量代謝通路激活對ALS體外細胞培養模型具有保護作用，初步說明該能量代謝通路可能參與ALS的發病。為進一步驗證課題組提出的ALS發病機制能量代謝假說，本課題擬在轉基因的ALS體外細胞培養、動物模型以及能量限制複合模型中，利用細胞生物學及分子生物學技術研究AMPK/SIRT1分子通路參與ALS疾病啟動與發展的分子機制，為ALS的診斷與病程評估提供新指標、為探索以調節能量代謝通路為靶點治療ALS提供理論依據。
