殷憲振

殷憲振

殷憲振,套用化學專業學士、計算機軟體與理論專業碩士、藥劑學專業博士,臨港實驗室研究員。具有豐富的影像學程式設計圖像處理數據分析經驗,及藥劑學、物理化學、成像光學、計算機科學等多學科專業背景,熟悉同步輻射X射線顯微成像、計算機斷層掃描螢光成像等多模態成像技術,自進入上海藥物研究所以來持續開展藥物製劑結構及生物醫學圖像數據挖掘相關的交叉學科研究工作。已發表SCI論文40篇,其中第一作者、通訊作者20餘篇;申請中國發明專利、國際PCT專利、軟體著作權12項,其中已獲授權9項。

於2009年開始在國內率先採用同步輻射光源顯微斷層掃描成像(SR-μCT)技術,與圖像處理、三維重構、三維建模相結合,系統開展在製劑結構相關方法學研究和製劑評價工作。發現具有特殊性質的製劑結構,經最佳化後設計籠狀片、動態結構滲透泵等新結構給藥系統,相關工作達到國際領先水平。

負責基於MOST/fMOST系統的全器官尺度亞細胞解析度影像平台建設,發展快速高精度的大樣品成像方法實現全腦高分辨圖譜採集及三維重構。開展醫學影像數據挖掘方法學研究工作,基於先進的三維可視化與定量表征方法,建立系統的神經系統、腦血管、脊髓血管、骨-腱組織、骨關節、肝臟血管的形態學數據分析流程。同時搭建自動化的處理能力達100TB/年的高效圖像處理系統。

科研進展

1、2023年8月2日,臨港實驗室殷憲振等人在Science Advances上發表了題為“Cross-scale tracing of nanoparticles and tumors at the single-cell level using the whole-lung atlas”的研究論文。本研究基於顯微光學斷層掃描/螢光顯微光學斷層掃描系統(MOST/fMOST),在三維空間、單細胞水平上,構建B16F10小鼠肺轉移瘤病理圖譜,闡明納米載體在腫瘤和臨近組織的時空分布規律,對其在腫瘤組織的滲透行為及靶向效率進行定量評價。本研究構建了一種全新的納米載體靶向效率的高精度可視化評估方法,突破現有腫瘤成像方法及瘤內納米載體可視化技術的瓶頸和局限性,為納米遞藥系統的設計和開發提供新的策略。
2、2023年8月8日,臨港實驗室殷憲振研究員等人受邀在ACS Molecular Pharmaceutics上發表了題為“Advances in Structure Pharmaceutics from Discovery to Evaluation and Design”的綜述當期封面文章。
3、2022年4月19日,臨港實驗室殷憲振研究員等人在Frontiers in Neuroscience雜誌上發表了題為“High-Resolution Digital Panorama of Multiple Structures in Whole Brain of Alzheimer's Disease Mice”的研究論文。該研究對基於顯微光學切片斷層成像(Micro-Optical Sectioning Tomography, MOST)系統獲得的全腦Nissl染色數據集,設計出針對相鄰灰度區間內多種結構元素的信息提取及重建策略,首次實現了基於Nissl染色的小鼠全腦Aβ斑塊分布可視化,在同一小鼠全腦中構建了Aβ斑塊及其周圍胞體、神經樹突、神經束和血管的高精度全景圖(圖1)。研究成果為深入理解AD相關病理狀態下的大腦解剖結構特徵提供了新思路。
