項目摘要

神經膠質細胞和神經元一起,是構成神經系統的基本細胞。膠質細胞不僅對神經元起營養和支持作用,而且通過分泌遞質主動調節神經信號傳遞。谷氨酸是最重要的興奮性神經遞質,也是膠質細胞分泌的主要遞質之一。關於膠質細胞分泌谷氨酸的機制(是否通過鈣離子介導的囊泡)問題有長期爭論。2005年我們套用電化學微電極和FM螢光成像發現了海馬星形膠質細胞釋放谷氨酸的一種新模式:鈣離子依賴性囊泡kiss-and-run(J Neurosci,2005;Neuroscientists, 2006),成為該領域研究熱點。為研究膠質細胞中谷氨酸囊泡分泌的調控機制,本申請將研究膠質細胞分泌囊泡的分子類型,以及已知兩種囊泡(溶酶體和20nm囊泡)被誘導釋放的動力學。這項研究的重要生理意義是,膠質細胞如何通過鈣離子依賴性谷氨酸囊泡釋放來主動調控神經信號傳遞。
