研究方向

1. 脂滴生物學
脂滴是重要的細胞器,存在於所有真核生物中。脂滴與其它細胞器互作,回響胞內外環境變化,動態調節細胞的脂類代謝和能量平衡,參與生物體的生長、發育、生殖、衰老、抗逆境等。脂滴在脂肪組織和非脂肪組織(如肝臟)中大量沉積,導致肥胖、脂肪肝、糖尿病等代謝性疾病。我們利用秀麗線蟲和小鼠等模式生物,結合細胞生物學、遺傳學、生物化學及分子生物學、代謝流等方法和技術,重點研究:1)脂類合成調控及其生物學功能;2)脂滴起源和增大機理;3)不同生理或病理條件下脂滴與其它細胞器互作維持能量平衡機理,進而揭示脂代謝紊亂相關疾病如脂肪肝和糖尿病等代謝性疾病的發病機理,並尋找新的疾病標誌物和治療靶標。
2. 脂肪肝發病機理及治療
肝臟是重要的代謝器官。臨床上,當肝臟中多於5%的肝細胞出現脂滴積累時,即為肝臟脂肪變性。隨著肝臟中脂肪過多的積累,脂毒性、胰島素抵抗等多種因素作用下,出現小葉炎症和肝細胞損傷等,肝臟脂肪變性發展為脂肪性肝炎;如果脂肪性肝炎中出現膠原沉積則發展形成肝臟纖維化,繼續惡化則導致肝硬化和肝癌。
建立了樂漏承境脂肪肝的細胞、小鼠、樹鼩和獼猴模型。發現樹鼩是一種新型的非肥胖型脂肪肝(non-obese fatty liver)動物模型,篩選到一些自發性肥胖、脂肪肝和糖尿病獼猴。基於上述脂肪肝細胞和動物模型,甩多屑利用轉錄組、蛋白質組、尋背和代謝組等,開展脂肪肝的發病機理研究,尋找新的疾病標誌物和治療靶標;同時開展減肥降脂藥物的篩選、評價和作用機理研究。
