科研成果
首次發現SUMO特異性蛋白酶SENP2基因缺失導致胚胎心臟發育缺陷,證實了SENP2在胚胎心臟發育中的調控作用,以及SUMO修飾調控PcG複合物的活性,進而調控機體發育和細胞分化的分子機制;首次發現SENP2在myostatin的信號通路及骨骼肌發育中的重要作用;首次發現了SENP2缺失引起鉀離子通道蛋白Kv7的過度SUMO修飾,從而降低Kv7的通道活性,引起顛癇和猝死,證實了SENP2對神經系統及其引起的心臟綜合症有重要影響。相關研究成果發表在《Neuron》(IF: 15.982),Molecular Cell (IF: 14.464)和《Journal of Biological Chemistry》(IF: 4.6)等國際一流刊物,並獲得MD Anderson腫瘤中心內科部基礎科學類“傑出論文獎”。研究成果得到了許多國際主流媒體的關注和報導,如ScienceDaily和EurekAlert等。近年來在Neuron,Molecular Cell,Journal of Biological Chemistry,Journal of Cellular Physiology,Toxicology等國內外學術刊物上發表論文20餘篇。同時,相關成果得到國家和陝西省自然科學基金面上項目的資助。

研究領域
類泛素修飾(SUMOylation)是一種新的蛋白質修飾形式,是影響其所修飾的蛋白質功能與定位的一個關鍵性調節機制。它參與調節多種細胞生命活動過程,包括細胞周期、細胞分裂、DNA損傷修復、細胞內轉運、調節蛋白穩定和基因轉錄等。SUMO修飾是一個動態可逆的過程,由SUMO特異的活化酶(E1)、結合酶(E2)和連線酶(E3)相繼作用來催化完成,其去SUMO修飾的過程(De-SUMOylation)則是由SUMO特異性蛋白酶家族(SENP)來完成。本實驗室主要研究蛋白質的SUMO修飾,特別是由SENP2介導的去SUMO修飾過程和對細胞信號轉導的調控機制,以及它們在發育與疾病過程中的作用與意義。主要研究領域有:
1. SENP2的靶蛋白分子與生物學功能
SENP2的功能是通過對靶蛋白質的去SUMO化,來改變靶蛋白的活性來實現的。因此,研究SENP2的生物學功能,首先要鑑定SENP2的靶蛋白分子。我們通過功能性篩選和基因敲除等策略來尋找和鑑定SENP2的靶蛋白質,並研究SENP2調控靶蛋白的作用與機制,從而來明確SENP2的生物學功能。
2.調控SENP2的細胞信號網路
SENP2的去SUMO化參與了許多細胞生命活性過程,也會受到細胞信號網路的調控。因此我們在鑑定SENP2靶蛋白的基礎上,研究細胞信號通路對這一去SUMO化過程的調節作用與機制,並依此建立SENP2的細胞信號調控網路。
3. SENP2在發育與疾病中的作用與意義
SUMO修飾的靶蛋白分子與發育和疾病有密切的關係,因此SENP2通過這些靶蛋白分子的去SUMO化修飾,必將參與對發育和疾病的調控。我們利用細胞、模式動物及疾病標本,綜合運用分子、細胞和發育生物學以及病理學的知識與手段,研究蛋白質的SUMO修飾在發育與疾病過程中的作用機制,為醫學實踐提供理論基礎。
