簡介

浙江大學醫學院閔軍霞教授團隊發現,心臟細胞“鐵死亡”是心臟疾病的關鍵致病機制,並基於分子調控機制提出了多種預防心臟損傷的有效途徑。這一發現為心肌病及心肌梗死等致死性拳勸請心臟疾病的防治提供了新策略,相關論文於2019年1月29日發表於《美國科學院院報》。
心肌細胞作為一種終末分化細胞,它的死亡將導致數量的減少,從而引起心臟結構和功能上的缺陷,加劇心力衰竭。”王福俤介紹說,細胞死亡方式主要分為探定堡經典的壞死和凋亡兩種,近年來隨著對細胞死亡機制的深入研究,鐵死亡等多種新的細胞死亡方式被相繼發現。
為了深入研究心台協棗髒疾病中細胞死亡的作用,研究團隊採用了多種細胞死亡類型抑制劑處理小鼠,發現抑制細胞鐵死亡能夠明顯改善心肌損傷,通過抑制劑來阻斷細胞鐵死亡還能夠明顯減輕由心肌缺血再灌注導致的急慢性心臟損傷
必需微量元素鐵與心臟疾病的關係廣泛而緊密。我們發現,攝入膳食鐵過多,特別是血紅素鐵,會顯著增加心血管疾病風險,而鐵死亡抑制劑、鐵螯合劑、線粒體抗氧化劑、低鐵膳食等是預防心臟損傷的有效途
“關於鐵死亡的研究還主要集中於體外細胞水平,本研究為我們深入了解鐵死亡在心臟損傷中的作用,探索利用鐵死亡抑制劑治療心臟損傷提供了大量的證據。”哈爾濱工業大學生命科學中心研究員高明輝評價說。
該研究提示鐵死亡可能參與臨床上心肌梗死等多種心臟疾病的發病,下一步應促進這一成果套用於臨床,為心臟病防治做出更多貢獻。

運用醫學及生命科學前沿技術以中國高發腫瘤患者多組學大數據結合功能篩選為出發點,探索適用於中國消化道惡性腫瘤霉求定霸基因型的有效新型靶向治療、免疫治療及克服靶向耐藥的綜合治療策略;基於中國長壽人群佇列系統性研究長壽的遺傳學、代謝及免疫特徵,為人類衰老及相關慢性疾病防治提供了新思路。
