項目摘要

胞吞與胞吐是細胞生長過程中最基本和重要的過程,同時也是一個多層次複雜的調控體系,其中細胞骨架在其過程中發揮著重要的調控作用,但是其具體的調控機制有待深入研究。本項目擬以植物單細胞極性生長模式系統- - 花粉管作為實驗體系,利用膜片鉗、細胞免疫標記、螢光分光光度計、電鏡等技術,結合酶解、快速冷凍及顯微注射等手段,對花粉管生長過程中頂端的胞吞/胞吐活性以及Ca2+相關指標進行檢測,定量分析細胞骨架解聚劑和穩定劑處理後,細胞骨架結構、質膜Ca2+通道活性、胞內Ca2+濃度以及胞吞/胞吐活性的動態變化規律。通過對上述各項觀測結果的綜合分析,進一步揭示細胞骨架對胞吞/胞吐的調控機理,同時也為深入探討高等植物有性生殖的分子機理提供理論依據。
