納米生物醫藥技術及其新藥成藥性。研究基於腫瘤微環境的、具有靶向給藥+聯合用藥+程式殺傷性能特徵的納米給藥系統,以期實現:①靶向遞送藥物至腫瘤組織部位—②進入腫瘤細胞微環境定量共同釋放藥物—③藥物協同作用並誘導細胞程式性凋亡。將該技術套用於難溶性藥物腫瘤靶向納米給藥系統,基因治療藥物納米給藥系統,抗腫瘤多藥耐藥納米給藥系統,並進行相關新藥的成藥性及生物學效應研究。
研究方向
1、難溶性藥物腫瘤靶向納米給藥系統:將被動靶向與主動靶向相結合:a)採用新型納米膠束載藥技術和新型藥用輔料,解決難溶性藥物給藥困難問題,並通過控制載藥納米粒尺寸大小,利用EPR被動靶向效應使載藥納米粒在腫瘤部位絮集; b)利用細胞表面腫瘤特異性受體,將其配體接枝修飾於載藥納米粒表面,增加藥物特異性主動靶向遞送轉運,以實現提高藥效和降低毒性作用目的。
2、小核酸藥物納米遞送系統:採用新型高分子材料載體和自組裝技術,設計構建小核酸藥物納米遞送系統,解決核酸藥物體內的高效靶向遞送、高效轉染與胞內功能表達等關鍵難題,促進小核酸等基因治療藥物技術的發展與套用。
3、抗腫瘤多藥耐藥靶向聯合用藥納米給藥系統:基於腫瘤靶向納米給藥技術,研究將具有抗腫瘤多藥耐藥和抗腫瘤作用的兩種不同藥理作用的藥物(化藥或核酸藥物)製成複方納米給藥系統製劑進行聯合用藥,靶向遞送複方藥物至腫瘤微環境,使其按確定比例在腫瘤細胞胞內微環境內同時釋放並發生藥理協同作用,實現顯著逆轉腫瘤多藥耐藥性,增加藥物敏感性和有效性。
