研究課題
1.酶抑制劑的設計、合成、生物活性及機理研究
在生物體中,幾乎所有的生化反應都是在酶催化下完成的,一旦酶的功能失常,就會導致疾病,因此常以酶為靶位來設計藥物。我們研究的酶有HIV逆轉錄酶、蛋白酪氨酸激酶和蛋白酪氨酸磷酸酯酶。我們根據酶的三維結構和酶與底物的作用特性,設計、合成一系列特定化合物,從中篩選出具有高效抑制活性的抑制劑。並通過NMR、質譜和X-射線晶體衍射分析等方法研究酶與抑制劑的相互作用機理。
2.生物有機反應及其機理研究
採用有機化學方法模擬生物化學反應過程,並探討相關反應機理;採用有機化學方法構建具有生物活性和(或)功能的生物有機分子,如活性肽、活性核苷、活性肽-核苷綴合物、活性肽-糖綴合物和具有生物標記功能的化合物等。
3.金屬催化體系的研究
設計、合成和篩選金屬催化劑(如銅、鎳催化劑)的配體,並與金屬催化劑一起構成催化體系,用於高效構建C-N、C-O、P-C和C-C鍵;設計、合成和篩選金屬催化劑(如銅、鎳、鈀、銠催化劑)的配體,並與金屬催化劑一起構成催化體系,用於不對稱有機化合物的合成。
4.有機催化劑的研究
設計、合成具有特定結構的有機化合物,用於催化有機化合物的不對稱合成。
