本研究結合項目組多年果梅種質資源和分子生物學的研究基礎,在採用石蠟切片找出雌蕊敗育關鍵時期的基礎上,擬通過高通量測序分離與果梅雌蕊敗育相關的microRNAs,預測其靶基因並通過螢光定量PCR、RLM-RACE以及轉基因進行表達分析和功能鑑定,初步篩選出與雌蕊敗育相關的關鍵microRNAs,然後利用原位雜交技術等對上述關鍵microRNAs進行細胞定位及其對AGAMOUS等雌蕊調控基因表達的影響,鑑定出果梅雌蕊敗育的關鍵microRNAs,初步明確microRNA在果梅雌蕊選擇性敗育中作用,為揭示果梅雌蕊敗育的分子機制提供依據。
