項目摘要
不同於體細胞,哺乳動物的卵母細胞具有極性,導致其獨特的不均等減數分裂。這種不均等分裂對於卵胞質內的營養和遺傳物質的保存是至關重要的,直接影響到後期的受精以及胚胎髮育。到現在為止,對於卵母細胞不均等分裂的分子機制了解的還很少。之前的工作證明微絲直接參與了卵母細胞的極性形成。而我們前期的研究證明微絲相關蛋白Arp2/3複合物參與了這一過程,但是,還不清楚G蛋白酶相關效應因子是否參與了哺乳動物卵子的不均等分裂。為了研究G蛋白酶相關因子在卵母細胞極性形成過程中的功能和機制,本項目將採用RNA干擾、過表達、基因敲除等技術手段,結合活細胞工作站觀察,深入研究G蛋白酶Cdc42效應因子LIMK和Cofilin在小鼠卵子紡錘體遷移和定位,極性產生、極體排出等過程中的功能,並闡明其作用機制和信號通路,進一步拓展對卵母細胞極性產生的認識,為卵子質量控制、受精及後期胚胎髮育的調控提供理論基礎。
