項目摘要
哺乳動物Toll樣受體(Toll like receptors,TLR)在宿主先天抗感染免疫中發揮重要作用,而鵝TLRs的分子特徵和抗病毒感染的免疫功能仍不清楚。本項目利用RT-PCR和RACE技術從鵝組織中克隆出TLRs的cDNA全序列,經生物信息學分析明確其分子結構並構建分子遺傳進化樹。利用基因表達定量和免疫組織化學染色,明確TLRs在健康鵝體及NGVEV感染鵝體內的細胞定位、組織表達譜及其因感染而發生變化的規律。進一步以原代鵝免疫細胞為模型,利用基因表達定量鑑定鵝TLRs的激動劑,並明確下游細胞因子的表達情況。在此基礎上,通過共轉染報告基因和目標基因、SiRNA基因沉默技術及免疫中和技術明確NGVEV與TLRs的識別、結合及其相互作用的分子機制。本研究將闡明鵝TLRs的生物學特性並明確其介導的宿主抵抗NGVEV感染的分子機制,研究結果將豐富水禽先天抗病毒免疫學基礎知識。
