項目摘要
大豆在發芽期和幼苗期對水淹澇害敏感,發掘耐淹基因資源可促進耐淹遺傳育種工作。申請人前期研究發現國外引種野生大豆PI342618B在發芽期和幼苗期具有強於栽培大豆的高耐淹特性。本申請擬針對該野生大豆比較研究其種子、幼苗適應淹水逆境的形態解剖、生理生化特點;利用PI342618B衍生的2個重組自交系群體和由染色體片段導入係為主構建的種質群體分別通過QTL連鎖定位和關聯分析揭示其耐淹性的遺傳構成;創建次級群體精細定位耐淹重要QTL並篩選候選基因,轉錄組與定量PCR分析相結合研究應答淹水逆境的基因調控系統;同時開發分子標記用於標記輔助轉育野生大豆耐淹基因,選育耐澇栽培新種質。研究預期揭示野生大豆對不同時期淹水逆境的特殊適應機制,明確其與栽培種質耐淹性的遺傳差異,並發掘優異基因用於育種實踐。這對深入了解野生和栽培大豆種質耐淹適應進化規律具有重要意義,也可為大豆耐淹分子育種奠定材料和方法基礎。
