項目摘要
痛覺是人體重要的保護性功能,慢性痛又是困擾人類的重大疾病之一。腦對痛覺信息加工的動態過程至今不明。本項目以傷害性雷射刺激作為人和動物的正常痛覺模型,以足底注射福馬林作為動物持續性病理痛模型,配合條件性和非條件性實驗設計,利用清醒自由活動大鼠神經元群單位放電多通道同步記錄技術,同步記錄丘腦內、外側核群、初級和次級軀體感覺皮層、前扣帶皮層、內側前額葉、後頂葉皮層及杏仁複合體在疼痛刺激期間的神經元群放電活動,分析中樞在正常及病理狀態下處理疼痛的感覺、情緒和認知信息時,上述腦區內部及其相互間的信息動態流動情況。並在正常人及慢性痛患者記錄雷射痛覺誘發電位,運用動態源分析技術,分析正常及病理疼痛發生時人腦不同區域活動及其相互間的動態信息流動過程,以期揭示編碼疼痛的感覺、情緒和認知信息的中樞過程和機制,以及此機制在病理情況下的異常改變,為治療慢性頑固性疼痛提供理論依據。具有重大的理論和實際意義。
