約翰克倫威爾生於俄亥俄州托萊多。1907年在克利夫蘭的劇團當演員，1911年任劇團導演，1912年在百老匯演出舞台劇《小婦人》。1928年進入電影界，在派拉蒙影片公司與人合作導演了有聲片《啞吧》等多部影片。

1944年，他執導的影片《自君別後》獲得該年最佳影片金像獎的提名，並當選為該年《每日電影》的十大佳片之一。戰後，他一度重回舞台演出，後重返好萊塢執導了《安娜與暹羅王》等多部影片，五十年代，由於與他訂有拍片契約的電影老闆懷疑他“左傾”，因而離開好萊塢，去百老匯任編導和演員。於1979年9月26日去世，終年九十二歲。
