項目摘要
針對梨生產中普遍存在果實風味品質較差的突出問題,我們開展了以下研究:(1)採用梨基因組重測序結果篩選基因組內均勻分布的SNP位點,構建梨果實糖、酸性狀的SNP基因晶片,實現利用基因晶片進行梨品種資源及雜種後代果實糖、酸性狀的基因分型;(2)建立利用轉錄組、蛋白質組分析及全基因組關聯分析等多種手段篩選梨果實糖、酸性狀重要候選基因的技術體系,完善轉基因番茄驗證重要候選基因功能的技術,挖掘出梨果實糖、酸性狀基因(基因家族);(3)明確梨果實糖、酸性狀相關的重要轉錄因子、轉座子和small RNA等,揭示果實糖、酸性狀形成的‘基因-基因’互作關係及基因調控網路,闡明梨果實糖、酸性狀形成的分子及調控機制。研究結果主要包括:首先,從梨中克隆了梨果實糖轉運蛋白PbSUT、PbHT、PbSOT、PbTMT基因,己糖激酶基因PbHXK、PbFRK和山梨醇代謝相關基因 PbS6PDH、PbSDH。分析了這些基因家族的結構和進化特徵,證明了PbSUT2促進轉基因番茄成熟果實中蔗糖含量的提高,而果糖、葡萄糖和可溶性總糖含量下降;PbHT1促進轉基因番茄成熟果實中己糖和可溶性總糖含量的積累,但對蔗糖含量無顯著影響;PbSOT2的表達、NAD-SDH與NADP-SDH 的活性和己糖的積累呈正相關。PbHXK1和PbFRK1負調控果實糖積累,並影響植株的生長發育。其次,對酸積累相關基因PbALMT9、PbTDT1和PbVHA-c4研究,發現5 個ALMT家族基因與蘋果酸代謝相關,其中PbALMT9不僅轉錄表達水平高於其他基因,且表達模式與梨果實整個發育時期的蘋果酸含量變化呈正相關;PbTDT1有助於果實發育過程中有機酸的積累,超表達轉基因番茄成熟果實中的蘋果酸、延胡索酸含量高於野生型和 RNAi轉基因株系,而檸檬酸含量低於野生型和 RNAi轉基因株系;PbVHA-c4促進梨果實中檸檬酸的積累,其超表達轉基因株系的番茄果實中檸檬酸含量明顯高於野生型。研究結果不僅為梨果實品質定向改良奠定理論基礎和提供基因資源,也為分子輔助育種提供有效方法。
