項目摘要

哺乳動物的胃液為極端酸性(pH 1-3),是防止食物細菌感染的一道天然屏障,但仍然有少數細菌(例如大腸桿菌)能夠適應這種酸脅迫。了解細菌的抗酸機制會加深我們對微生物生理和致病機理的理解,具有重要的科學意義和醫學價值。本實驗室近幾年在揭示大腸桿菌膜間質抗酸系統作用機制方面取得了一定的進展,尤其是最近發現分子伴侶蛋白HdeA和雙功能分子伴侶-蛋白酶DegP協同作用促進酸變性的蛋白復性,從而有效提高了細菌抗酸能力。本項目擬進一步研究這種協同抗酸的機制。我們將運用非天然胺基酸體內交聯法鑑定HdeA和DegP在活細胞中的天然底物蛋白,並揭示DegP作為分子伴侶蛋白促進HdeA的天然底物蛋白復性的機制;另外,我們也將研究DegP作為蛋白酶發揮抗酸功能的作用機制。本項目的研究成果將為我們理解革蘭氏陰性細菌(以大腸桿菌為代表)膜間質抗酸系統的普遍機制提供重要的依據和見解。
