項目摘要
六六六(HCH)是一種曾經廣泛使用的長殘留殺蟲劑。它有α、β、γ和δ四種異構體,其中γ-HCH與α-HCH的微生物降解途徑已被闡明。目前,δ-HCH的微生物降解是HCH降解領域的研究熱點。研究δ-HCH的降解途徑及相關基因對於全面了解六六六微生物降解機制具有重要意義。本課題組已在δ-HCH微生物降解方面取得了一些進展。分離到一株能高效降解六六六四種異構體的菌株Sphingomonas sp. BHC-A,研究了其降解δ-HCH的部分途徑。本項目擬進一步深入研究該菌株對δ-TDOL和δ-2,3,5-TCDL(已證明的BHC-A菌株降解δ-HCH的中間代謝產物)的降解途徑,最終推導出完整的δ-HCH降解途徑。並通過轉座子隨機插入突變和SEFA-PCR相結合的方法克隆出降解途徑中的關鍵酶基因。預期結果有助於闡明微生物降解δ-HCH的分子機理,並為微生物修復δ-HCH污染環境提供理論依據。
