卵母細胞體外成熟(in vitro maturation,IVM)技術在輔助生殖領域,特別是PCOS和腫瘤患者,已展現出重要的套用潛力,但是IVM低效率限制了其臨床套用。IVM卵母細胞核質發育不同步是其效率低下的核心因素,因此探討促進卵母細胞胞質成熟的分子機制和解決新策略,將有助於推動其臨床套用。Kisspeptins 是近年發現的一組下丘腦肽,在卵母細胞體內成熟及排卵過程中,通過調節線粒體功能及改善絲裂原活化蛋白激酶(MAPK)活性,在卵母細胞成熟過程中起關鍵作用。本研究利用小鼠IVM模型,通過體外添加Kisspeptin54,從分子,亞細胞,細胞,胚胎及成體水平,探討其對IVM卵母細胞發育潛能的改善及其使用對子代的長期安全性和風險性,並從線粒體功能及MAPK調控角度,對Kisspeptin54促進IVM卵母細胞發育的分子機制進行詮釋。研究成果為IVM效率改善及臨床推廣提供理論基礎。
