項目摘要

外周敏化,即初級感覺神經元的過度興奮與骨癌痛的發生直接相關;KCNQ/M通道對神經元的興奮性起到剎車閘樣調節作用,可以抑制神經元興奮性的過度增強,提示該通道在骨癌痛的發生中可能具有重要作用。癌症微環境可以產生大量的NGF,參與癌症痛的形成;NGF可以通過酪氨酸磷酸化與PIP2水解來抑制M通道的功能、提高神經元的興奮性。我們前期的工作發現,骨癌大鼠DRG神經元上M通道的表達和功能明顯抑制,這種抑制可能與DRG神經元興奮性的增加和骨癌痛的發生有關。本課題擬從以下三個方面闡明KCNQ/M通道在骨癌痛中的作用及其機制:①建立大鼠骨癌痛模型;②是否由於KCNQ/M通道的抑制而引起DRG神經元的過度興奮,進而引起外周敏化和骨癌痛的發生?③是否由NGF通過酪氨酸磷酸化與PIP2水解來抑制KCNQ/M通道,從而引起DRG神經元過度興奮,導致外周敏化和骨癌痛的發生?旨在闡明癌症痛的發病機理,尋找治療新靶點。
