項目摘要
廣泛存在於生物體內的HtrA蛋白質家族是一類重要的熱休克蛋白,同時具有分子伴侶和蛋白酶的雙重功能,在生物體對蛋白質進行質量控制中起關鍵作用。人類的HtrA同源蛋白質與各種疾病如癌症、帕金森症、老年痴呆症都有關係。HtrA在大腸桿菌中被稱作DegP,對於細菌外膜蛋白的生物合成和質量控制至關重要。我們擬以DegP為對象,採用單分子探測技術研究HtrA家族的分子伴侶和蛋白酶雙重功能的分子機制。將單分子探測這一物理化學新方法引入相關研究,可以揭示出新的現象和規律,增進人們對生物體實現蛋白質的正確摺疊、抵禦外部脅迫環境、清除錯誤摺疊的蛋白質的機理的了解,為人類認識相關疾病的發病機理和提出治療對策提供科學依據。這是一個有相當難度的課題,但我們經過多年的探索與準備,已經具備了實現預定目標的研究基礎,可以圓滿完成任務。
