魏澤文

魏澤文,男,北京理工大學生物醫學工程系副教授(研究員)/ 博士生導師,微電子機械系統方向。主要研究微電子機械系統在生物醫學工程領域的套用,側重將前沿MEMS技術套用於生物醫療領域,包括利用基於矽微加工技術的微納器件和基於軟光刻的微流控器件對蛋白、核酸、細胞及組織進行精細分析及操控。發表SCI及本領域頂級會議論文40餘篇,多為JCR一區頂刊,H因子14,申請專利十餘項,出版書籍一章節。

科學研究

研究課題

微電子機械系統(Micro-Electro-Mechanical System)及其在生物醫學工程領域的套用(Bio-MEMS)。微電子機械系統(MEMS)是在微電子技術的基礎上發展起來的,融合了矽微加工,並套用現代信息技術構成的微型系統。它包括感知和控制外界信息(力、熱、光、磁、化)的感測器和執行器,以及進行信號處理的電路。從國防到工業控制,從醫療設備到消費電子,MEMS技術都有廣泛的套用。本人的研究領域是將前沿MEMS技術套用於生物醫療領域,包括利用基於矽微加工技術的微納器件和基於軟光刻的微流控器件對蛋白、核酸、細胞及組織進行精細分析及操控。

研究方向

(1)液體活檢、稀有細胞檢測及單細胞操控、分析和測序
探索多種前沿的微結構、微器件和生物分析方法,實現對細胞(及其行為)、蛋白與核酸的精確操控、篩選和分析,用於多種疾病的體外診斷。例如:
首次將循環腫瘤細胞檢測的所有步驟(包括血液處理、細胞分離、鑑定、計數及單細胞分析和基因擴增)都集成在單塊微流控晶片上,結合基於深度學習的圖像分析算法,實現了在3.5小時內處理2毫升全血並無需人工干預,在近200例臨床試驗中獲得了迄今為止最優的性能。該系列技術有望成為癌症液體活檢及癌症單細胞測序的有效工具。
(2)基於單細胞操控的單克隆抗體系列新技術
基於多種新穎的微結構及單細胞測序技術,在單克隆抗體生成、篩選、測序領域開發了一系列新方法和新技術。
(3)基於細胞電穿孔(Electroporation)的核酸輸運
探索了多種微電極結構,全面提升了向體外細胞或活體組織輸運核酸的效率和通量。用於體外細胞的系列成果已轉讓形成產品,性能顯著優於國外同類產品。用於活體組織的系列成果已進入DNA疫苗等治療領域,例如向恆河猴遞送針對HIV的DNA疫苗,已取得優秀的免疫回響(抗體滴度比現有金標準痘病毒遞送高60%)和保護效果。該系列技術不僅能夠顯著提高電穿孔實驗技術的效率和通量,還有望使電穿孔技術成為提升核酸藥物療效的有力工具。
