項目摘要
結構可控的DNA/RNA 複合物是開發安全、高效的非病毒類基因傳遞載體的首要條件。DNA/RNA在生理條件下攜帶有大量負電荷,可歸屬為聚電解質。計算機模擬結果表明,在電場存在下,聚電解質的構象會發生由線團到棒狀的轉變,而且抗衡離子的分布也會發生變化。這都會影響聚電解質的複合能力及所形成的複合物結構。本項目基於這一原理,提出利用毛細管電泳、微流體晶片電泳和板電泳來製備受電場調製的DNA/RNA 複合物;利用散射、TEM等物理手段來確定複合物結構和形態;並進一步建立複合物結構與傳遞效率之間的關係。本項目從聚電解質的基礎理論出發,引入電場這一能量因素,研究以DNA/RNA為代表的生物分子在非平衡態的複合規律,對製備高效傳遞載體有重要意義,對於透徹理解生命過程也有重要價值。
