隋廣超教授,博士生導師。北京大學本科及博士學位,瑞典Karolinska 醫學院醫學博士學位,美國哈佛大學醫學院博士後。在美國Wake Forest 大學醫學院做9年的獨立PI,至副教授,於2014年全職引進東北林業大學生命科學學院。在Cell, PNAS, Oncogene, Nucleic Acids Res, Mol Cancer (第一或通訊作者) 等刊物共發表80餘篇SCI文章,英文專著一部。在美國主持過NIH R01, 美國癌症協會和美國國防部等多項癌症研究基金。自2014年回國後,主持了三項國家自然科學基金面上項目和一項黑龍江省國自然基金重點項目。指導學生獲得國際基因工程大賽(iGEM)金獎5次,銀獎3次,單項獎3次;全國生命科學聯賽一等獎兩次,二等獎一次。在美國和中國多次參加基金評審。發明shRNA抑制基因表達方法,該方法已被世界範圍實驗室廣泛使用。獲兩項發明專利(美國和中國各一項)。研究領域為癌症發病的分子機制,抑癌藥物開發和天然活性物質的抑癌機理研究。

研究領域
1.藥用植物抑癌活性成分的藥效機理研究:藥用植物中的很多成分具有抑癌活性,但其藥效機理不明,限制其臨床套用。為突破該瓶頸,我們將探究植物藥用成分在肝癌細胞內的分子靶點、所影響的重要基因及細胞生物途徑,創立科學的聯合給藥方式以有效抑制肝癌發展並降低對機體副作用。首次在細胞水平驗證了鞣花酸聯合阿黴素或聯合順鉑的抗肝癌活性,並在小鼠模型中鑑定了鞣花酸聯合阿黴素使用,可以顯著加強阿黴素抗肝癌活性,減弱由高濃度下阿黴素引發的心肌毒性。此外,我們還研究了百里香琨,地榆皂苷II的植物天然活性物質的抑癌活性。
2.發明了以DNA載體為基礎的RNA干擾(RNAi)技術(即shRNA方法)。這一技術的發明為許多生物學科的研究提供了全新的方法,尤其是對基因和蛋白質功能的研究具有重大的創新性,也進了RNAi技術在醫療上的套用。這一方法已經成為世界範圍實驗室用於研究基因功能的常規實驗技術。該方法的DNA載體成為Ambion/Life Technologies公司的第一代產品(商品名pSilencer)。做為第一作者的相關PNAS文章已被引用上千次。
3.利用對Yin Yang 1(YY1)蛋白在腫瘤發生中功能的研究來探索幾種常見癌症(乳腺癌,前列腺癌,肺癌)的致病機制。我的多年的研究成果確立了YY1的癌基因地位。作為一個多功能蛋白,YY1在癌症發生中的終極作用一直有爭議。我在2004年發表的Cell文章中發現了YY1對p53的負調控,奠定了YY1的癌基因的地位。這些研究進一步確定YY1的癌基因地位,並揭示了YY1在癌症治療中作為靶基因的可能性。通過干擾YY1與癌蛋白之間相互作用以抑制腫瘤生長的多肽已被授權專利。
4.對其它抑癌基因、原癌基因及非編碼RNA的研究:(1)利用基因晶片等方法對抑癌基因SOX7的研究,發現其上、下游調控因子及可能的調節機制;(2)對DMTF1研究,發現了這一基因產生mRNA的不同選擇性剪接(alternative splicing)形態對乳腺癌發生和發展具有完全相反的作用,以及調節該可變剪接的分子機制;(3)研究了幾種microRNA對原癌基因EZH2、YY1和FASN的調節作用;(4)利用多個乳腺癌及它們對應的癌旁組織樣品中的長非編碼RNA(lncRNA)的分析,發現了多種在乳腺癌中差異性表達的lncRNA,目前正在對它們的生物學意義和對乳腺癌的調節功能進行鑑定。
