項目摘要
生命個體的發育主要取決於遺傳基因DNA序列,但其複雜性和多樣性不能僅由生物學中心法則解釋。在生命個體的發育過程中生物大分子的化學修飾對決定細胞命運起著關鍵的調控作用。具有遺傳性的生物大分子化學修飾的研究是當前生命科學研究的一個前沿課題。本申請的研究內容是酶對核酸(DNA和RNA)進行化學修飾和去修飾的動力學機理。由於以前研究手段的制約,關於核酸化學修飾動力學機理的研究尚處在初期。我們擬將實驗(單分子探測)和理論(分子動力學模擬)結合在一起,採用先進的研究手段深入探究酶對核酸進行化學修飾的動力學過程,以便清楚地了解酶如何和以多快的速率發現被修飾和去修飾的位點,以及酶在發現位點後對其進行化學修飾的分子機制和反應動力學。這些研究將揭示相關生物分子反應的化學本質,有助於更深刻地認識相應的生物過程。
