研究概述
RNA分子在蛋白質翻譯、RNA修飾和加工、基因調控等多種過程中發揮著重要的作用。葉克窮研究員實驗室的主要興趣集中在和RNA生物學相關的結構研究,包括真核生物核糖體的組裝、RNA修飾和RNA沉默等領域,主要使用X射線晶體學、電鏡、核磁共振、生化、酵母遺傳操作等技術手段。
1. 真核生物核糖體組裝
蛋白質工廠——核糖體是由RNA和蛋白質組成的超級複合物,它在體內的組裝需要經歷高度複雜的過程。在出芽酵母Saccharomyces cerevisiae中,有200多個保守的蛋白質組裝因子和大量非編碼RNA參與核糖體的組裝。我們以出芽酵母為模式生物,研究核糖體體內組裝的分子過程、組裝因子的結構和功能以及核糖體前體的結構。
2. H/ACA和C/D RNA蛋白質複合物
H/ACA 和 C/D RNA是兩大類古老的非編碼RNA,它們和蛋白質結合形成多元複合物,主要在RNA的假尿嘧啶合成和2'-O甲基化修飾中,發揮結合特異底物的嚮導作用。我們研究H/ACA和C/D 兩類RNA蛋白質複合物的空間結構、生化機制和生物發生過程。
3. RNA沉默
多種小RNA(siRNA, miRNA, crRNA)作為特異性決定因子在真核和原核生物中控制著基因的沉默。我們研究這些小RNA的加工、調控機制和效應複合物。
