項目摘要

齲齒病是累及全人類的細菌感染性疾病,WHO已將其列入危害人類健康的三大疾病僅次於心腦血管病和癌症,我國衛生部和國家教委已把齲齒列為2000年學生重點防治疾病之一。大量研究證明變形鏈球菌是最主要的致齲細菌,對其進行大規模蛋白結構研究將深化對變鏈菌致齲分子機理的理解,有助於尋找齲病防治的有效靶標以及可作疫苗的抗原蛋白。經多年努力建成的北京大學結構基因組學平台可進行每年1000個以上基因克隆及表達,能夠勝任變鏈菌的全基因組蛋白結構研究,並在2004年進行了200多個變鏈菌基因的先導研究。在此基礎上,擬進行(1)變鏈菌全基因組非膜蛋白基因約1500個的大規模質粒構建;(2)高通量表達、純化可溶性蛋白;(3)培養蛋白晶體,對衍射好的晶體進行結構解析;(4)在結構基礎上,對未知功能的蛋白進行深入分析,由結構信息推動功能研究
