同年9月赴羅切斯特大學醫學中心(University of Rochester Medical Center)從事博士後研究工作,2014年9月通過中國科學院“百人計畫”引進回昆明動物研究所工作。現為神經系統疾病課題組負責人。長期從事於精神分裂症遺傳和神經發育機制研究,在精神分裂症遺傳機制研究方面取得顯著成績。首次發現和鑑別了一些新的精神分裂症易感基因如CAMKK2和ZNF323,探討了這些基因在精神分裂症發生中的可能機理。同時還揭示了Gata3和Lmo4基因在聽神經系統發育中的重要作用。已在Molecular Psychiatry、The American Journal of Psychiatry、Schizophrenia Bulletin、Human Molecular Genetics、The Journal of Neuroscience、Journal of Medical Genetics等雜誌發表論文二十餘篇。

研究方向

精神分裂症是一種影響患者思維、情感和行為的嚴重的神經系統疾病。作為人類最常見的疾病之一,精神分裂症影響了大約1%的人口。由於病因複雜,反覆發作以及大多在青壯年發病,精神分裂症嚴重影響患者生活,同時也給患者家屬和社會帶來了沉重的負擔。目前的研究表明遺傳因素和環境因素在精神分裂症的發生中都有作用,其中又以遺傳因素為主(精神分裂症遺傳力達到0.8左右)。
實驗室主要圍繞精神分裂症的遺傳機制和發病機理進行深入的系統研究。結合人類遺傳學、生物信息學以及神經生物學,從分子、細胞、模式動物和人等多個層次進行重要精神分裂症易感基因的功能及其機制研究,主要目標是闡明精神分裂症遺傳機制和致病機理,最終為預防和治療精神分裂症提供診斷及預後的分子標誌物以及治療靶標。目前研究興趣集中在探索和發現新的精神分裂症易感基因,並深入研究這些精神分裂症易感基因如何影響神經系統發育功能,最終導致精神分裂症發生。目前研究方向主要集中在以下三個方面:
1.利用人類遺傳學和整合生物信息學研究精神分裂症的遺傳機制
雖然精神分裂症的遺傳學研究已經取得顯著進展,然而精神分裂症遺傳基礎仍不清楚。精神分裂症是多基因疾病,多個微效基因的累加效應結合環境因素共同作用導致精神分裂症發生。目前的人類遺傳學研究(包括遺傳連鎖、相關、GWAS、外顯子測序等)已經鑑別到大量的精神分裂症易感基因,然而這些基因僅僅只能解釋精神分裂症一小部分遺傳力(heritability),暗示著大量效應較小的精神分裂症易感基因仍有待進一步的發現。利用人類遺傳學和整合生物信息學,本課題組將系統性分析和鑑別新的精神分裂症易感基因。
2.運用小鼠和靈長類動物模型研究精神分裂症的致病機理:
目前精神分裂症的病因仍不清楚,累積的研究表明精神分裂症是由神經系統發育異常導致的。 因此,了解精神分裂症易感基因如何影響神經系統發育對了解精神分裂症發生至關重要。本課題組利用基因敲除方法在小鼠靈長類動物模型中深入研究精神分裂症易感基因在神經系統發育中的作用,探索精神分裂症易感基因的分子功能和致病機理。
3.探索精神分裂症的潛在治療靶標和開發抗精神疾病藥物
依託昆明動物所豐富的靈長類資源,本課題組將嘗試建立精神分裂症靈長類動物模型,並利用靈長類動物模型探索精神分裂症的潛在治療靶標和開發抗精神疾病藥物。
