項目摘要

活性氧(ROS)是一類化學性質非常活潑的含氧分子或自由基,在細胞信號轉導、能量代謝以及許多病理過程中都發揮至關重要的作用。線粒體是ROS生成、清除、平衡調控的一個主要細胞器。申請人最新研究發明了一種超氧陰離子螢光蛋白探針cpYFP,並發現了新的線粒體超氧生成事件超氧炫(Superoxide Flash)。但有關線粒體超氧炫的機理、功能及疾病相關性等方面的許多重要問題亟待解答,目前世界各地100多家實驗室正在進行跟蹤研究,共同催生一個嶄新的領域。基於前沿動態和核心優勢,本項目擬利用生物物理學、生物信息學、分子生物學以及生理和病理學等手段,研究超氧炫信號的產生與調控的分子機制,尋找超氧炫所激活的下游信號分子和信號通路,並試圖確定介導超氧炫發生的線粒體膜通透性孔道(mPTP)的分子實質。此外,通過發展ROS在體分子成像技術,探討超氧炫線上粒體能量代謝以及代謝相關重大疾病中的作用與意義。
