項目摘要

鼻腔是禽流感病毒入侵動物的主要途徑,鼻腔免疫後可阻斷禽流感病毒進入機體。鼻腔黏膜對滅活禽流感病毒抗原的吸收和樹突狀細胞有效攝取抗原是限制滅活病毒鼻腔免疫效率的兩個主要瓶頸。樹突狀細胞伸出突起通過上皮細胞連線攝取抗原呈遞給淋巴細胞是啟動黏膜免疫的關鍵。因此,本項目主要從促進滅活病毒的吸收和誘導鼻腔黏膜樹突狀細胞的分化和成熟提高鼻腔局部和全身免疫反應的效率。首先進行樹突狀細胞的分離和培養,然後建立呼吸道上皮細胞模型calu-3細胞與樹突狀細胞共同培養體系,研究影響共同培養模型中樹突狀細胞攝取滅活禽流感病毒的因素,最後套用體外試驗(細胞共培養模型)和體內(小鼠和雛雞)試驗一起證明CpG、膽酸鈉配合滅活禽流感病毒對樹突狀細胞分化和成熟、鼻腔局部細胞因子、局部Toll樣受體以及局部和全身血清中特異性抗體的影響。本研究結果將為滅活病毒進行鼻腔免疫提供充分的理論依據和預防禽流感發生提供一條簡便的免疫途徑
