項目摘要
在國家自然科學基金項目人和小鼠睪丸晶片差異基因庫的生物信息學分析與表達研究中，發現一新的睪丸特異性基因TSG23。該基因僅在出生後15日齡的小鼠睪丸組織表達，且呈年齡依賴性增加；在無精子症患者睪丸組織的表達明顯減少或消失。.本項目：1、採用原位雜交技術，分析TSG23在小鼠和人睪丸生精細胞中的亞細胞定位；2、採用免疫組織化學方法，進一步確定TSG23在小鼠和人睪丸組織的表達特徵；3、採用基因敲除技術，研究TSG23在小鼠精子發生過程中的作用及其機理；4、篩選TSG23表達減少或消失的無精子症患者，通過基因組DNA測序分析，確定患者TSG23基因的突變、缺失或單核苷酸多態性。該項目將為進一步探索精子發生分子機理提供新的依據，也可能為男性不育症的治療和計畫生育新藥開發提供新靶點。
