項目摘要

Transient receptor potential (TRP) channels是一個非選擇性陽離子通道家族,它們是人和動物的分子感覺器,分別感受溫度、痛覺、機械、味覺等基本環境因子和傷害性刺激。當TRP被激活時, 鈣離子便流入TRP離子通道,並通過鈣離子依賴性(激酶,分泌,收縮)產生細胞功能。因此TRP鈣離子通透性是一個重要問題。本項目研究的重點是使用周專-Neher發明的測量陽離子通道鈣離子分電流的方法(Pf,例如NMDAR通道Pf=10%),研究TRPV1特性並測定迄今未知的溫度敏感TRPV2離子通道的Pf,從而為研究它們的生理功能研究提供重要基礎。申請人曾參與周專實驗室測定HCN通道的Pf, 並掌握了Pf方法(Yu et al, PNAS,2004;Am J Physiol, 2007)。我們的預實驗結果已經初步檢測了TRPV1的鈣離子Pf=5.4%。
