在對梨自交不親和性生理及分子機理研究的基礎上,綜合套用雷射細胞生物學及生物化學等的研究手段和技術,從花粉管骨架、超微結構及生理生化代謝等方面深入研究梨花柱S-RNase介導花粉管程式性死亡的機制。內容包括花柱S-RNase介導自花花粉管程式性死亡過程的花粉管骨架(微絲及微管)、細胞核、液泡、線粒體、高爾基體等超微結構變化特點,花粉管核DNA的凝聚與片段化特性,花柱S-RNase與花粉管凋亡誘導因子(AIF)、類Caspase、細胞色素C、VPE等的協同作用,花柱S-RNase介導的花粉管囊泡轉運動態(胞吞/胞吐)及分布特性等。將闡明花粉管感受花柱S-RNase的'毒害'而凋亡的'應答'反應特點,揭示配子體型自交不親和性反應中花柱S-RNase介導自花花粉管程式性死亡的細胞超微結構與生化代謝機制,完善薔薇科植物自交不親和性機理的理論體系,有重要學術理論和潛在套用價值。
