以陸地棉徐州142和無長絨無短絨突變體(fuzless-lintless, fl)為實驗材料,通過大規模分離、鑑定和研究棉花纖維伸長期相關基因,尋找用於遺傳工程改良棉纖維的靶基因。選用開花後10天(10DPA, 10 days post anthesis)、棉花纖維伸長速度最快的陸地棉纖維與fl突變體胚珠作為起始材料,用PCR-Select cDNA Subtraction方法獲得了纖維特異的cDNA減法庫,分離到172個纖維特異或高表達的基因,為闡明棉花纖維發育的分子機制奠定了良好的基礎。
