項目摘要
動脈粥樣硬化是最常見的血管疾病,人體內磷酯信號小分子S1P合成代謝的異常對動脈粥樣硬化的發生髮展以及心血管的健康產生重要影響。本項目以揭示有機磷酸酯類物質對磷脂合成代謝影響機制及其毒理效應作為研究目標,通過建立血液樣品中磷脂S1P代謝合成相關物質和有機磷酸酯類物質的監測方法,評價人群有機磷酸酯類物質的暴露水平對人體磷脂S1P水平的影響,並結合小鼠暴露實驗弄清有機磷酸酯對磷脂S1P合成代謝的影響;研究有機磷酸酯類物質與PPARγ、磷脂S1P受體的結合活性和結構特徵,從細胞水平上闡明有機磷酸酯類物質對磷脂S1P合成代謝和對泡沫細胞形成和內皮細胞粘附功能的影響及其機制,為該類物質的風險評價提供毒理學基礎。
