項目摘要
神經管缺陷(NTDs)是一類常見致死、致殘性先天缺陷,是影響出生人口素質的主要原因之一。NTDs的病因及發病機制尚未完全清楚。本課題組前期研究提示,多環芳烴(PAHs)可能與NTDs發病有關。本項目結合人體研究和動物模型,研究PAHs的致NTDs作用,檢驗氧化應激是否為PAHs的致畸機制。人體研究採用病例對照研究方法。病例為引產NTDs胎兒,對照為引產無缺陷胎兒。檢測肝組織內PAHs水平;中樞神經組織內氧化-抗氧化狀態、PAH-DNA加合物、生物大分子氧化損傷標誌、PAX3和p53基因及蛋白表達水平,及細胞凋亡水平,分析PAHs與NTDs的關係及氧化應激所起作用。通過孕鼠體內染毒和體外全胚胎培養,構建PAHs致NTDs動物模型,檢測胎鼠中樞神經組織內氧化-抗氧化狀態、DNA加合物、生物大分子氧化損傷標誌、Pax3和p53基因及蛋白表達水平,及細胞凋亡水平,揭示PAHs致NTDs分子機制。
