項目摘要
穀物澱粉是微米尺度的多糖聚合體,分子量巨大、構型複雜,澱粉整體的晶體學結構目前未解。本課組在前期研究中發現穀物澱粉存在納米尺度的中間體,在研究澱粉中間體分子量的MALDI-TOF質譜測定中,又發現各種多糖峰以常態分配組合、分子量呈現有序排列,顯示出多糖片段和封閉結構的有序規律。本課題針對中間體MALDI-TOF質譜實驗顯示出多糖組成的有序規律,引進同步輻射的X射線衍射和小角X射線散射實驗技術,分析、測定玉米澱粉中間體的多糖 直鏈片段和封閉結構等有序組成規律與晶體結構之間的關係;使同步輻射的X光束線站在解決澱粉中間體、多糖聚合問題中的套用打開突破口,使澱粉晶體學研究獲得新的認知和進展;在其他分析方法的配合下,為優異穀物種質的性狀、專用食品原料,提供納米尺度的數據和指標;為保障食品質量和安全、指導有效使用糧食資源,提供參考依據和實驗技術。
