項目摘要
腫瘤分子顯像和分子靶向治療是診治腫瘤的重要研究內容,顯像的目的是為了更好地治療。最新研究表明,腫瘤的發生伴隨著多個基因位點突變,靶向下游的某單一靶點的治療效果很可能是有限的。在腫瘤分子靶向治療面臨巨大挑戰的時候,腫瘤放射靶向治療更突顯其優勢。為了使放射性核素標記的多肽分子探針具有更加有效的放射治療效果,目前多採取多肽分子多聚化的辦法提高腫瘤對放射性分子探針的攝取並延長其在腫瘤的滯留時間。但是單純多聚化其負面效應也是非常嚴重的,正常組織特別是腎臟對放射性分子探針的攝取明顯提高。因此單純的多聚化是不切實際的,必須對多肽分子進行結構改造。本課題針對腫瘤表達的integrin αvβ3設計以RGD多肽為靶向分子的放射性分子探針,通過多肽分子結構改造最佳化出具有優良體內外性質的111In標記的顯像探針,並在此基礎上用90Y和177Lu標記性質優良的靶向分子,進行腫瘤放射靶向治療的相關研究。
