項目摘要
馬鏈球菌獸疫亞種(SEZ)舊稱獸疫鏈球菌,是豬鏈球菌病的主要病原。其可引起豬的腦炎,但機制不明。本研究以SEZ強毒菌株ATCC35246為研究對象,利用轉座子誘變技術,建立轉座子突變體庫,以豬腦微血管內皮細胞(PBMEC)為血腦屏障模型,篩選SEZ與突破血腦屏障模型相關的毒力因子,發現粘附蛋白,並通過小鼠模型評價其功能;構建PBMEC文庫,利用酵母雙雜交技術,從文庫中篩選與SEZ突破血腦屏障相關毒力因子的互作蛋白,並進行鑑定,尋找相關粘附受體;同時通過SEZ對PBMEC的侵襲實驗,評價SEZ產生的溶血素、鏈激酶和PBMEC金屬蛋白酶表達量的變化對血腦屏障模型通透性的影響。在此基礎上,採用細胞信號通路發現晶片分析蛋白互作等引起PBMEC細胞信號激活的途徑。結果通過闡明SEZ突破PBMEC血腦屏障模型的分子機制,能進一步解析SEZ致豬腦炎的機理,並為相應的藥物及疫苗設計提供依據。
