摘要

從廣東 省陽江, 電白和韶關等地55-95'C的溫泉中採集水樣, 經 分離得到21株G-, 不產生芽孢的高溫菌. 它們都能產生 L-乳酸脫氫酶. 根據表觀特徵, DNA的同源性和G+Cmol%等 特徵可歸入棲熱菌屬 (Thermus) . 但對葡萄糖不產酸和 不液化明膠等表型特徵不同於已發表的種. 又根據該群內 的DNA/DNA雜交結果, 有高的同源性 (大多在80%以上) , 而與已發表的種僅有40.5-70.7%的同源性. 結果表明可成 立一獨立的類群, 建議為棲熱菌屬的一個新種, 因具不液 化明膠的特性, 命名為非解朊棲熱菌 (Thermus nonproteolytics Cai & Yang sp. nov.).以菌株 HG-42為模式株, 保藏於中國科學院微生物研究所.
Abstract A new species of obligately anaerobic, sporeforming, thermophilic, cellulolytic, saccharolytic, proteolytic Clostridium is described.Two strains(Hll, H12) of this species are isolated from silage (grass) on milk cow field.
Cells in PY-cellobiose broth culture are Gram-negative rods, 0.3 to 0.6 by 1.5 to 5.5 micrometer, motile and peritrichous, occur singly or in pairs,occasionally in short chains. Long filaments may be seen in older cultures. Spores are oval or round, terminal and subterminal, swell the cell.Sporulation occurs readily in PY-cellobiose broth or cellulose broth after incubation for 24 hours. Free spores are round, have an exosporium and appendages.Deep colonies in PY-cellobiose agar roll tubes are lenticular, watery, yellowish color, 1-2mm in diameter, with entire margins.In PY-cellulose agar roll tubes they are pin-like, less than 1mm in diameter, raised, colorless, translucent, with an irregular margins.After incubation for 5 days, colonies are surrounded by clear zones, representing cellulose hydrolysis, that usually are 3 to 7 mm in diameter.Cultures in PY-cellobiose broth are moderate turbid with a stringy sediment and have a pH of 5.0 afterincubation for 5 days. The optimum temperature for growth is 50-53'C. There is little growth at 37'C, and none at 65'C.The optimum pH for growth is 7.5. No growth in media without fermentable carbohydrates. In a chemically defined medium, growth factors required arebiotin, pyridoxine, vitamin B12, and p-aminobenzoic acid. Fermentation products (per 100 mL of culture) are 1.7-2.4 meq of succinic acid,1.0-1.2 meq of acetic acid, less than 1 meq of ethanol, hydrogen, and carbon dioxide in 1% PY-cellobiose or PY-cellulose medium.The mol % G+C of the DNA is 38.8 (Tm). Other characteristics of the species are given in Table 1.
The above characteristics show that the species differ from the known close species of genus Clostridium, sach asClostridium stercorarium. and other cellulolytic thermophile: Clostridium thermocellum, Clostridium thermolacticum, Clostridium josuiand Clostridium thermocoprial. The name Clostridium succinogenum sp. nov. is proposed for this species, referring to chief product in nonvolatile acidof the species. The type strain (H11) has been deposited in the General Microbiological Center of China Committee of Culture Collection of Microorganism as AS 1.1782.
