項目摘要

雞球蟲病由艾美耳屬的7種球蟲感染引起,分別寄生於十二指腸和小腸上段、中段、下段及盲腸,具有明顯的寄生部位特異性。寄生部位特異性取決於侵入部位特異性,但決定部位特異性的蟲體配體和細胞表面受體尚不完全明了。本項目擬選取不同部位寄生的代表性蟲體,根據蟲體配體與細胞表面受體相結合的原理,提取蟲體子孢子膜蛋白或全蟲可溶性蛋白,與雞不同腸段上皮細胞結合,提取結合後細胞膜蛋白或總蛋白,進行2-D電泳分析,並用子孢子蛋白高免血清識別,選取識別蛋白點,通過質譜分析,確認蟲體配體分子。再觀察配體重組蛋白抗血清阻斷蟲體侵入和配體重組蛋白對攻蟲的免疫保護作用,確定蟲體配體。在此基礎上,用酵母雙雜交確認細胞受體分子,用抗血清被動免疫保護試驗進一步驗證受體功能,並觀察受體在整個腸道的分布,從而確定雞球虫部位特異性的關鍵分子。對闡明雞球虫部位特異性的分子機理有重要意義,對研究頂復門其他蟲體的部位特異性也有重要借鑑意義。
