研究方向

病原細菌/人體微生物組與宿主互作-以效應蛋白為中心
微生物與其宿主之間的相互作用是生物物種間最複雜的相互作用之一。一方面,在與人類或者其它動植物宿主億萬年共同進化的過程中,共棲微生物(Commensal)成功適應了宿主環境,並對宿主的發育、免疫及神經系統都產生重要的影響;另一方面,病原菌(Pathogens)則能突破宿主的免疫屏障,實現感染。雖然抗生素和疫苗的發明使我們能對付大多數的病原細菌(Pathogenic bacteria),但是,病原細菌也逐漸發展出了抵抗抗生素的策略,出現了超級抗性菌,使病原細菌特別是耐藥性細菌又一次成為人類的重大威脅;同時,病原細菌還發展出了精巧複雜的毒力系統,以適應宿主體內環境、躲避或克服宿主的免疫體系,導致各種感染症甚至癌症等,其中最重要的毒力系統就是細菌的分泌系統(Secretion System)。通過相當於細菌“武器”的分泌系統,細菌可以向宿主體內分泌數量眾多、功能各異的毒力蛋白-效應蛋白(Effector proteins,也就是細菌的“子彈”),效應蛋白與宿主細胞表面或宿主細胞中的各種靶點蛋白相互作用,改變宿主的各種信號傳導途徑如細胞凋亡、自噬和細胞周期等,從而適應宿主環境,克服宿主免疫系統的攻擊,達到讓細菌自身在宿主內增殖的目的。
研究微生物和宿主相互作用的學科稱為細胞微生物學(Cellular Microbiology),細菌效應蛋白功能的闡明是該學科研究的重點。(1)闡明效應蛋白的生化功能,可以讓我們更深入理解病原細菌致病以及適應宿主環境的分子機制;(2)以這些效應蛋白為工具,發現宿主固有免疫(Innate Immunity)調控的新途徑,是免疫學的新領域;(3)效應蛋白本身還可以作為抗菌藥物作用的理想靶位點,研發針對這些效應蛋白的活性先導物(相當於鈍化或收繳細菌的“子彈”),有可能獲得避免耐藥性產生的新型抗生素;(4)可以運用合成生物學(Synthetic Biology)策略在微生物細胞、癌細胞或免疫細胞中構建效應蛋白調控迴路(Circuit) ,實現對疾病相關細胞活性的人工調控。
圍繞以上方向,我們的研究興趣具體如下(詳見主頁中的“研究興趣”欄目):
(1)效應蛋白的生化功能及宿主免疫反應機制以目前臨床上典型的胞內病原細菌嗜肺軍團菌(Legionella pneumophila)、結核分枝桿菌(Mycobacterium tuberculosis)以及重要的植物病原細菌茄科勞爾氏菌(Ralstonia solanacearum)為模型,重點研究其致病的關鍵效應蛋白的表達調控及其生化功能,闡明這些蛋白與宿主靶蛋白相互作用進而調整宿主免疫系統的分子機制。
(2)靶向效應蛋白的抗結核藥物研製結核分枝桿菌是結核病的病原,感染了我國近三分之一的人口,是國人健康的重大威脅,急需新的抗結核藥物。我們以結核分枝桿菌關鍵效應蛋白為靶點,建立相應的篩選系統,從微生物代謝產物中篩選效應蛋白的抑制劑,並研究其作用機制,期望獲得新型的、能避免耐藥產生的新型抗結核藥物。
(3)腸道微生物組與宿主的互作及其分子機制腸道菌群是腸道內寄居的種類繁多的微生物的總稱。腸道微生物是代謝的重要參與者,同時代謝產生的脂肪酸等促進腸道上皮細胞生長與分化。腸道菌群之間可以通過如分泌系統分泌的效應蛋白和群感效應(Quorum sensing)等維持菌群的平衡。腸道菌群失調(dysbiosis)會增加疾病的易感性,包括胃腸道疾病、代謝類疾病(如肥胖、糖尿病等)、免疫類疾病、精神類疾病和癌症等。對腸道微生物組與疾病關係的研究,將為開發基於微生物的疾病診斷、疾病干預、治療和益生菌產品的研發等提供基礎。我們綜合運用基因組學、代謝組學和生物信息學技術,研究腸道微生物組(microbiome)或腸道細菌及其效應蛋白和代謝產物與炎症和癌症等的發生、發展的關聯性及其分子機制;同時,運用合成生物學的相關策略,建立重要腸道菌的分子遺傳作業系統,以期實現對腸道細菌功能的精準調控。
