項目摘要
多糖是自然界含量最豐富的生物聚合物, 廣泛表達於微生物、植物和動物中, 並以糖基化的方式形成糖蛋白參與所有蛋白質分子的結構穩態和功能調節,主要包括細胞信號傳遞、基因表達、免疫回響以及蛋白質摺疊等方面,蛋白質糖基化的變異是生物體發生變異的主要途徑之一,準確高靈敏度測定蛋白質糖基化變化是理解和調節生命行為的基礎工作。本課題擬以建立蛋白質糖基化特徵譜高靈敏度分析方法為目標,通過新型免疫識別體系-新衍生方法-同位素標記衍生試劑相結合,開發出基於毛細管電泳-雷射誘導螢光高靈敏度檢測及毛細管電泳-無鞘液接口質譜鑑定的糖基化變異檢測的新方法,為蛋白質糖基化的快速鑑定特別是疾病前期預警建立有效工具。
